| 摘 要： | 目的研究遗传性视网膜色素变性大鼠(Royal College of Surgeon's rat,RCS rat)在视网膜变性发展过程中是否有内源性视网膜干细胞(retinal stem cells,RSCs)激活.方法RCS-p (视网膜含色素的变性大鼠)按出生后视网膜变性的发展状况分为变性早期(RCS 15 d)、中期(RCS 30 d)、晚期(RCS 90 d)3个时相点,分别采用相应时期的RCS-rdy p (视网膜含色素的正常大鼠)作为对照组(control groups,C15 d,C30 d,C90 d),每组4只8眼,对各组视网膜切片进行免疫荧光组织化学(抗Chx10)实验,以鉴定视网膜干细胞;进一步采用Western blot检测各组视网膜神经上皮总蛋白中Chx10表达.结果①各正常组及变性15 d大鼠视网膜各层抗Chx10免疫荧光检测均为阴性,变性中、晚期视网膜光感受细胞层部位Chx10免疫荧光阳性;②Western blot检测各变性组视网膜神经上皮总蛋白中Chx1O均有表达,但变性中期Chx10表达显著高于早期和晚期(P＜0.05).结论 RCS大鼠变性过程中视网膜感光细胞层出现视网膜干细胞激活.
