摘    要:目的探讨复合壳多糖人工皮肤移植家兔后表皮层角蛋白的表达以了解移植后皮肤表皮的分化情况.方法分别将复合壳多糖人工皮肤移植前、移植后1个月、移植后2个月、正常人皮肤、正常兔皮肤石蜡块切片,用抗广谱角蛋白多抗、抗角蛋白K5和K10单抗进行免疫组化染色,观察表皮层角蛋白的表达和表皮分化情况.结果复合壳多糖人工皮肤移植后1个月及2个月表皮层均有较强的角蛋白表达,与正常皮肤相似;基底层以上细胞有角蛋白K10表达,且越接近表皮表面表达越强,与正常人皮肤相似,基底层细胞有角蛋白K5表达,但明显弱于正常人皮肤.结论复合壳多糖人工皮肤移植家兔后表皮层均有角蛋白表达,有增生性细胞和分化细胞,表皮分化良好、层次清楚,与正常皮肤相近.

Abstract:Objective To observe the expression of epidermal keratin and the epidermal differentiation after graft of composite chitosan artificial skin onto experimental rabbits. Methods Immunohistochemical staining of the paraffin embedded sections from composite chitosan artificial skin before grafting, one month, and two months after grafting, and from normal human skin and normal rabbit skin was performed using anti broad spectrum polyclonal antibody to keratin, basal cells and suprabasal cells with monoclonal antibodies to cytokeratin K5 and cytokeratin K10, respectively. The epidermal keratin expression and the epidermal differentiation were observed. Results The keratinocytes in the composite chitosan artificial skin expressed keratin one month and two months after grafting, similar to the normal epidermal keratinocytes. The suprabasal cells expressed cytokeratin K10 one month and two months after grafting. The closer to the surface of epidermis, the stronger the expression was, similar to the normal human epidermis. The basal cells expressed cytokeratin K5 one month and two months after grafting, but the expression was obviously weaker than that of the normal human skin. Conclusion Keratin expression, proliferative cells, differentiated cells, and good differentiation of epidermis with clear layers are found in the epidermis after grafting of composite chitosan artificial skin onto rabbits. The epidermal reconstruction of composite chitosan artificial skin after grafting onto rabbits is satisfactory. The composite chitosan artificial skin is similar to the normal human skin.
