| Abstract: | Objective To observe the dynamic changes of proinflammatory cytokines IL-1β,IL-6 and anti-inflammatory cytokines IL-10 in LPS-induced RAW 264.7 cells and observe the effects of cholecystokinin octapeptide(CCK-8) on their expression.Methods After RAW 264.7 cells were stimulated with 1 mg/L LPS for 0,3,6,12,24 and 48 h respectively,the expression peak times of IL-1β,IL-6 and IL-10 were selected to investigate the effects of CCK-8 at the doses of 10-10,10-9,10-8,10-7 and 10-6 mol/L respectively.Cytokines protein and mRNA expressions were assayed by ELISA and RT-PCR.Results The expression of IL-1β,IL-6 and IL-10 mRNA and protein were induced by LPS in RAW 264.7 cells in a time-dependent manner.CCK-8 inhibited LPS-induced IL-1β and IL-6 expression and augmented IL-10 expression in a dose-dependent manner.When the cells were treated for 24 h,CCK-8 augmentation resulted in higher LPS-induced IL-10 expression when compared with that after 48 hours’ treatment.Conclusion CCK-8 may participate in the anti-inflammatory response by inhibiting the expression of proinflammatory cytokines IL-1β and IL-6,meanwhile augmenting the expression of anti-inflammatory cytokine IL-10 induced by LPS. |
