异位和正常子宫内膜间质细胞对上皮细胞间隙连接细胞间通讯的影响

摘    要:目的 研究体外异位和正常子宫内膜间质细胞对内膜上皮细胞间隙连接细胞间通讯(gap junctional intercellular communication, GJIC)功能的不同影响.方法 取24例卵巢处子宫内膜异位灶以及10例正常子宫内膜,分离、纯化上皮细胞和间质细胞,建立体外上皮细胞单独培养、上皮细胞与间质细胞共培养模型.在激光共聚焦显微镜下采用荧光漂白后恢复技术分别检测各组上皮细胞的GJIC功能.结果 ①异位内膜上皮、间质细胞纯度分别为92%、95%,对照组内膜上皮、间质细胞纯度分别为95%、98%;②单独培养的异位内膜上皮细胞与单独培养的正常内膜上皮细胞的GJIC功能无显著性差异(P>0.05);③与正常间质细胞共培养后,上皮细胞中GJIC功能明显上调,而与异位内膜间质细胞共培养的上皮细胞中GJIC功能未见明显变化.结论 异位间质细胞丧失了调节上皮细胞GJIC的能力,这种调节能力的异常可能与子宫内膜异位症发生有关.

Abstract:Objective To explore the different influences of ectopic and normal endometrial stromal cells on gap junctional intercellular communication (GJIC) in endometrial epithelial cells in vitro. Methods The epithelial and stromal cells were isolated and purified from samples including 24 endometriotic tissues located in ovaries and 30 normal endometria. The model of epithelial cells cultured alone and the model of endometrial epithelial cells cocultured with endometrial stromal cells were established in vitro. Laser scanning confocal microscopy (LSCM) was applied to determine the functions of GJIC in epithelial cells among different groups by fluorescence recovery after photobleaching (FRAP). Results The purity of epithelial and stromal cells was 92%, 95% in ectopic endometrial group respectively, and was 95%, 98% in control group respectively. No significant difference was found on GJIC between ectopic endometrial epithelial cells cultured alone and normal endometrial epithelial cells cultured alone (P>0.05). GJIC was up-regulated in epithelial cells cocultured with control endometrial stromal cells, but not in the epithelial cells cocultured with ectopic endometrial stromal cells. Conclusion That ectopic endometrial stromal cells lost ability to regulate GJIC in endometrial epithelial cells may be related to the pathogenesis of endometriosis.
