IL-10基因修饰对骨髓间充质干细胞体外免疫效应的影响

目的：研究IL-10基因修饰的骨髓间充质干细胞（BM-MSCs）体外免疫效应。方法:RT-PCR克隆大鼠IL-10基因并构建逆转录病毒pLNCX-IL-10，转染大鼠BM-MSCs；流式细胞术检测IL-10基因修饰的BM-MSCs（BM-MSCsIL-10）体外对T淋巴细胞亚群的影响，CCK-8法检测BM-MSCsIL-10及其培养上清液对体外混合淋巴细胞增殖的影响。结果:BM-MSCsIL-10能明显减少CD4 T，降低CD4 /CD8 比值，比BM-MSCs作用明显（P<0.05）；BM-MSCsIL-10及其培养上清液比BM-MSCs更能明显抑制T淋巴细胞增殖反应（P<0.05）。结论:IL-10基因修饰能增强BM-MSCs的体外免疫抑制作用。

Influence of IL-10 modification on in vitro immunosuppressive effect of bone marrow mesenchymal stem cells

Objective: To study the in vitro immunosuppressive effect of IL-10 modified bone marrow mesenchymal stem cells(BM-MSCsIL-10). Method: IL-10 was cloned by RT-PCR and was used to construct pLNCX-IL-10. The constructed retrovirus was transfected into rat BM-MSCs; T lymphocyte subsets in vitro influenced by BM-MSCsIL-10 were detected by flow cytometry. BM-MSCsIL-10 or its culture supernatant on in vitro mixed heterologous T lymphocyte proliferation were detected by CCK-8. Results: BM-MSCsIL-10 significantly reduced CD4 T and lowered the ratio of CD4 /CD8 more than BM-MSCs (P <0.05); BM-MSCsIL-10 and BM-MSCsIL-10 or its culture supernatant significantly inhibited T lymphocyte proliferation more than BM-MSCs (P <0.05). Conclusion: IL-10 modification can enhance the in vitro immunosuppressive effect of BM-MSCs.
