淫羊藿总黄酮对金属离子刺激单核细胞分泌肿瘤坏死因子的影响

摘    要:目的通过观察淫羊藿总黄酮(total flavones of Epimedium,TFE)对金属离子(Co^2+)刺激单核细胞分泌肿瘤坏死因子-α(TNF—α)的影响,探讨TFE抑制TNF—α分泌的机制以及防治关节假体的无菌性松动的可能性。方法采集36例健康志愿者的外周血,分离单核细胞,分组培养。实验分成6组。A组:仅单核细胞;B组:单核细胞+金属离子;C组:单核细胞+金属离子+帕米膦酸钠10mg/L;D组:单核细胞+金属离子+TFE 5mg/L;E组:单核细胞+金属离子+TFE 10mg/L;F组:单核细胞+金属离子+TFE 20mg/L。单核细胞培养48h后,ELISA法测定细胞上清液中TNF—α的含量。结果B组TNF-α的含量明显高于A组(P〈0.01);C、D、E、F组TNF-α的含量分别明显低于B组(P〈0.01);D、E组之间及E、F组之间TNF—α的含量差异均有显著性(P〈0.05)。结论金属离子可刺激单核细胞使其分泌溶骨性因子TNF—α的量增加。TFE与帕米膦酸钠作用效果相似,可降低溶骨性细胞因子TNF—α的分泌,随着TFE浓度的增加,其降低TNF-α分泌量的作用加强,从而有望成为防治关节假体松动的药物之一。

Abstract:Objective By observing the effect of total flavones of Epimedium (TFE) on secreting TNF - α due to stimulation of human monocytes by metal ion cobalt( Co^2+ ) , to discuss the role of TFE in decreasing the secretion of TNF -α, providing a theoretical support for using the herbal medicine to prevent and treat aseptic loosening of joint prosthesis. Methods Blood samples obtained from 30 healthy human donors were processed to get the monoeytes to perform the present experiment in 6 groups, group A: containing monocytes only; group B: monoeytes + metal ion ; group C: mono- cytes + metal ion + pamidronate 10 mg/L; group D: monocytes + metal ion + TFE 5 mg/L; group E: monocytes + metal ion + TFE 10 rag/L; and grou F: monocytes + metal ion + TFE 20 mg/L. The mixtures were cultured ford8 h to determine the concentration of TNF - α by ELISA. Results The level of TNF - α was much higher in group B than in group A, C, D, E and F ( P 〈 0.01 ). There was significant statistical difference between group D and group E ( P 〈 0.05 ) and between group E and group F (P 〈 0.05 ). Conclusion Being stimulated by metal ion, monocytes can secrete TNF - α.The inhibitory effect of TFE is similar to that of pamidronate, and is proportional to the concentration of TFE. It seems that TFE can be a potential therapeutic agent for the prevention and treatment of aseptic loosening of joint prosthesis.
