摘    要:应用平滑肌肌球蛋白轻链激酶(MLCK)体系,观察了重金属离子Cd~(2+)和Hg~(2+)对MLCK活性的影响,得到以下结果:1.Cd~(2+)和Hg~(2+)对MLCK具有双相作用,低浓度下激活MLCK,高浓度下抑制MLCK;2.Cd~(2+)对MLCK的激活作用,被钙调素拮抗剂氯丙嗪所抑制;3.疏基化合物能逆转Cd~(2+)和Hg~(2+)对MLCK的抑制。结果提示,Cd~(2+)和Hg~(2+)能通过活化caM使MLCK激活,高浓度时直接作用于MLCK抑制其活性。

Abstract:Cd2 + and Hg2+ were found to mumic the stimulatory action of Ca2 + on calmodulin-dependent myosin light chain kinase ( MLCK ) from chicken gizzards. Hg2 + , however, was less effective. The effect induced by Cda+ and Hg2+ were biphasic, stimulatory at low concentrations and inhibitory at high concentrations. The activation by Cd2+ of MLCK was inhibited by, chlorpro-mazine, a calmodulin antagonist. These facts may help to explain mechanism of heavy metal toxicity and the vascular toxicity of Cd2 + .
