Calbindin-D-28k对MPTP致小鼠黑质凋亡相关蛋白Bax表达的影响

摘    要:目的探讨在1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)致小鼠黑质多巴胺(DA)能细胞发生凋亡时,calb ind in-D-28k(CB)抗细胞凋亡的作用机制。方法MPTP连续5天腹腔注射构建中脑黑质DA能细胞损伤的小鼠模型,模型鼠脑黑质致密部立体定位注射携带CB基因的高滴度慢病毒颗粒(CB-H IV-Ⅰ),W estern b lotting方法检测CB的在体过表达以及鼠脑黑质部位凋亡相关蛋白Bax的表达变化。结果与空白对照组(control)和黑质定位注射空病毒颗粒组(H IV-Ⅰ)相比,注射CB-H IV-Ⅰ的实验组的黑质中CB的表达量显著升高(P〈0.05),而Bax的表达量明显降低(P〈0.05)。结论病毒颗粒CB-H IV-Ⅰ携带的CB基因在黑质细胞中获得了高效的表达;凋亡相关蛋白Bax可能参与了CB对黑质DA能神经细胞的保护作用。

Abstract:Objective To investigate the anti - apoptotic mechanism of calbindin - D - 28k (CB) in the MPTP - induced apoptosis of dopaminergic (DA) neurons in midbrain substantia nigra in mice. Methods MPTP was injected into the peritoneal cavity in 5 consecutive days to establish an injury model of DA neurons in the midbrain substantia nigra in mice, and high titer lentivirus particles carrying CB ( CB - HIV - Ⅰ ) were injected into the midbrain substantia nigra by stereotaxic apparatus. Western blotting was used to analyze the expression levels of CB and apoptosis - related protein Bax in substantia nigra, respectively. Results The expression level of CB increased ( P 〈 0.05), while that of Bax decreased in CB - HIV - Ⅰ groups compared with control groups and HIV - Ⅰ groups (P 〈 0.05). Conclusion The lentivirus particles of CB - HIV - Ⅰ were highly expressed in substantia nigra cells ; apoptosis - related protein Bax might participate in the protective effect of CB on DA neurons in substantia nigra.
