成年大鼠脑内具有神经生发功能的脑室下区细胞核三维数字化形态观察

摘    要:目的 研究成年大鼠脑内具有神经生发功能的侧脑室外侧壁脑室下区(subventricular zone, SVZ)和室管膜(ependymal layer, EL)各种细胞的形态学特征及其空间排布规律.方法 采用半薄切片染色技术和计算机三维重建技术,利用3D Slicer工具软件构建SVZ-EL区各细胞核三维表面模型,并建立相应的数据库.结果 ①在EL,主要是体积较大且呈球形的成熟室管膜细胞核;另有体积更大的球形细胞核贴近室管膜细胞深方;此外,还有体积较小而形状不规则的细胞核散在镶嵌于室管膜细胞之间.②在SVZ,可见特异性的同型细胞核聚集现象,此类细胞核体积较大且都呈椭圆体,具有迁移中成神经细胞核特征,故称之为成神经样细胞核.它们有的邻近EL、有的邻近纹状体,其周围的细胞核分属不同类型;另有散在分布的大而似球体的未分化细胞特征性细胞核,它们与聚集的成神经样细胞核间无特定空间位置关系.此外,还有大量散在分布的中、小型不规则细胞核.结论 在SVZ-EL区含有不同类型细胞核,其中成神经样细胞核呈聚集分布,但其周围没有特定结构包裹现象.此外,本研究实现了成年哺乳动物脑内神经生发区SVZ细胞核形态及其空间位置的数字化,可作为进一步研究SVZ神经生发的基础.

Abstract:Objective To investigate the morphology and spatial organization of the cells which have the function of neurogenesis and lie in the subventricular zone(SVZ) and ependymal layer(EL) of the lateral wall of the lateral ventricle in adult rat brain.?Methods The techniques of semi-thin sections and crystal violet staining and the computer technique using three-dimensional(3D) Slicer software were employed to reconstruct the 3D surface model of the various-shaped cell nuclei in the SVZ-EL.The nuclear image messages were digitalized and stored in the computer to form a database available for further study in this field.?Results(1) In the EL,the majority were the big global nuclei of the mature ependymal cells.A few bigger nuclei were located deep to the ependymal cells,and some small irregular-shape nuclei were scattered and wedged between ependymal cells.(2) In the SVZ,there were clusters of special homotypic nuclei,which were relatively big in size and elliptical in shape,and were named as neuroblast-like nuclei in the present study,for they resembled the migrating neuroblasts in appearance.The clustering nunclei were found near the EL or adjacent to the striatum,with the surrounding nuclei belonging to various types.There also scattered some big spheroid undifferentiated nuclei,which had no special spatial relationship with clustering neuroblast-like ones.Besides,there were still a lot of scattered nuclei in the SVZ,which were middle-to-small in size and irregular in shape.?Conclusions The nuclei in the SVZ-EL are of different types.Among them,the neuroblast-like nuclei are distributed in clusters,but the clusters are not enwrapped by any special structure.A digitalized study on the morphology and spatial organization of the nuclei in the neurogenetic SVZ of adult mammalian brain is accomplished in the present study,providing a basis for further investigation in this field.
