不同剂量链脲佐菌素诱导SD大鼠糖尿病肾病模型的研究

摘    要:目的 探讨不同剂量链脲佐菌素(SIZ)致SD大鼠糖尿病。肾病模型状况的差异。方法 58只雄性SD大鼠随机分为5组,模型组大鼠(4组,每组12只)腹腔一次性注射40、50、60、70ms/ks的STZ以建立糖尿病模型;非糖尿病对照组10只,仅予注射缓冲液。注射后2,4、72h检测血糖,8周观察糖尿病大鼠尿微量白蛋白、血尿素氮、肾脏指数及肾组织光镜和超微结构等变化。结果 腹腔注射STZ60mg/kg成模率高,模型死亡率较低。8周后大鼠尿微量白蛋白水平即升高,。肾脏超微结构检查系膜细胞增生、基质增多、肾小球基底膜增厚。结论 腹腔注射STZ60mg/kg是致糖尿病SD大鼠发生肾病的最佳剂量。

Abstract:Objective To investigate the differences in diabetic nephropathy model rats induced by streptozotocin(STZ) of varied dosage.Methods 58 male SD rats were randomly divided into 4 diabetic groups(n=12 each) and 1 control group(n=10) to receive varied doses of intraperitoneal STZ and buffer solution respectively. The blood sugar and general conditions were monitored 1-3 days after the injection.Eight weeks later,the urinary microalbumin,blood urea nitrogen,kidney index and the ultrastructure of kidney were examined.Results The success rate of model making was relatively high and the mortality was relatively low in the STZ 60 mg/kg group.After 8 weeks,the urinary microalbumin was found significantly higger than normal.Ultrastructural study of the kidney showed proliferation of mesangial cells,increase of mesangial matrix and thickening of glomerular basement membrane.Conclusion STZ 60 mg/kg ip is the dose of choice for inducing nephropathy in rat models of diabetes.
