壳聚糖/葡甘聚糖共混膜的生物降解性能初步研究

摘    要:目的观察不同比例壳聚糖和葡甘聚糖共混膜在体外及体内的生物降解性能.方法流延法制备壳聚糖与葡甘聚糖含量比例分别为3:0、2:1、1:1、1:2的混合膜.将其分别置于含/和不含溶菌酶的林格氏液及昆明鼠股部肌肉内,检测其在体外和体内的降解性能.结果壳聚糖/葡甘聚糖含量为3:0、2:1、1:1和1:2的混合膜在体外有溶菌酶存在时70d降解率分别为40.75%、65.7%、74.2%和84.6%;纯壳聚糖膜在体内降解时间为16周左右,壳聚糖/葡甘聚糖混合膜的降解时间为8~12周左右.结论壳聚糖/葡甘聚糖共混膜的降解性较单独的壳聚糖膜有了改善,可作为一种潜在的牙周诱导再生膜材料.

Abstract:Objective To study the degradation of chitosan/konjac glucomannan blend film in vitro and vivo.Method Blend films containing chitosan and konjac glucomannan solutions with different proportion of 3:0,2:1,1:1,1:2 were prepared by solvent-casting method.The in vitro degradation tests of the prepared blend films were conducted by incubating the films in Ringer's solution with or without lysozyme.The in vivo degradation tests of the prepared blend films were conducted by incubating the films in muscle of thigh of Kunming mouse.The degradation profiles were expressed as the accumulated weight losses of the films.Their degradation rate in vitro and in vivo were measured.Results The degradation rate of chitosan/konjac glucomannan blend films with different proportion(3:0,2:1,1:1,1:2)in Ringer's solution with lysozyme in 70 days was 40.75%,65.7%,74.2% and 84.6% respectively.The degradation time of chitosan/konjac glucomannan blend films in vivo was about 8 to 12 weeks,while the degradation time of chitosan films was about 16 weeks.Conclusion The degradation of the chitosan/Konjac glucomannan blend films is faster than chitosan film.The blend films can be used in periodontal tissue engineering scaffold.
