组织芯片技术检测肿瘤相关抗原在膀胱癌、癌旁组织中的表达

摘    要:目的利用组织芯片技术检测人膀胱移行细胞癌(bladder transitional cell carcinoma,BTCC)组织、癌旁组织中肿瘤相关抗原(tumor-associated antigen,TAA)的表达.方法含40块不同病理分级的人膀胱癌和40块癌旁组织组合的组织芯片一张,含64块不同病理分级的人BTCC组织芯片一张.免疫组织化学Elivision法检测膀胱癌组织和癌旁组织中TAA的表达,并对不同病理分级BTCC组织中TAA的阳性表达率进行比较.结果膀胱癌TAA的阳性染色主要位于细胞膜,BTCC组织中的总表达率为72.5%(29/40),癌旁组织中的总表达率为30%(12/40),差异有统计学意义(P〈0.05).96块不同病理分级的BTCC组织中,TAA的阳性表达率分别是:G164.3%(9/14)、G269.8%(30/43),G374.4%(29/39).两两比较,TAA在G1、G2和G3中的阳性表达无差异性,P〉0.05,差异无统计学意义.结论膀胱癌组织中TAA的阳性表达率显著高于癌旁组织,TAA的表达和BTCC病理分级无关.组织芯片技术是检测膀胱癌TAA的一种有效方法,具有高效、快捷的优点.

Abstract:Objective To detect the expressions of tumor-associated antigen (TAA) in human bladder transitional cell carcinoma (BTCC) tissues and adjacent tissues by tissue microarray assay. Methods One tissue chip contained 40 tissue masses of human bladder tumor at different pathological grades and 40 adjacent tissue masses, and the other contained 64 tissue masses of human BTCC at different pathological grades. Expression of TAA in bladder cancer and adjacent tissues were identified by using Elivisions immunohistochemical essay. The positive rate of TAA expression was compared among different pathological grades of BTCC. Results The positive staining of TAA mainly located in the membranes of bladder carcinoma cell. The total expression rate of TAA in bladder.tumor was 72.5% (29/40) , while 30.0% (12/40) in adjacent tissues. The difference was statistically significant (P 〈 0.05). In 96 tissue masses of BTCC at different pathological grades, the positive expression rate of TAA was respectively 64.3%(9/14) in G1, 69.8% (30/43) in G2 and 74.4%(29/39) in G3. Multiple comparison showed the positive expression of TAA had no statistical difference between G1, G2 and G3 (P 〉 0.05). Conclusions The positive expression of TAA in Bladder Cancer is higher than that in adjacent tissues. There is no correlation between the positivity expression of TAA and the pathological grade of BTCC (P 〉 0.05). Tissue microarray technology is an effective way for detecting TAA of BTCC, and this method has the merits of high efficiency and facility.
