玉屏风散生物转化液对正常小鼠抗氧化功能及免疫抑制小鼠T细胞亚群、IL-4和IFN-γ的影响

摘    要:目的利用米曲霉菌株(AS3.951)对玉屏风散进行生物转化,观察玉屏风散生物转化液对正常小鼠抗氧化功能及免疫抑制小鼠免疫功能的影响。方法测定正常小鼠外周血和肝组织中SOD、GSH-Px酶活性、MDA含量;运用流式细胞仪测定环磷酰胺所致免疫抑制小鼠外周血和脾CD4^+T和CD8^+T亚群的百分比及其比值;用ELISA法测定环磷酰胺所致免疫抑制小鼠外周血中IL-4和IFN-γ的含量。结果玉屏风散生物转化液组SOD、GSH-Px酶活性高于对照组和玉屏风散煎剂组(P〈0.01)、MDA生成量低于对照组和玉屏风散煎剂组(P〈0.01);玉屏风散生物转化液组CD4^+T比例、CD8^+T亚群比例、CD4^+T/CD8^+T,IFN-γ的含量均高于环磷酰胺组(P〈0.01,P〈0.05),IL-4含量无明显差异;玉屏风散生物转化液组CD4+T亚群比例、CD4^+T/CD8^+T,IFN-γ的含量均高于玉屏风散煎剂组(P〈0.01),CD8^+T亚群比例无明显差异,IL-4含量无明显差异。结论经米曲霉(AS3.951)生物转化的玉屏风散比玉屏风散煎剂更能提高正常小鼠抗氧化功能,更能提高免疫抑制小鼠的免疫功能。

Effects of Yupingfengsan-transform-product on T-lymphocyes and IFN-γ and IL-4 of cyclosphamide-immunosuppressant mice and the antioxidant function of normal mice

Abstract:Objective To study the effects of Yupingfengsan--transform-- product on blood the-- dependent T--lymphocyes and blood IFN--γ and IL--4 of eyclosphamide--immunosuppressant mice and the antioxidant function of normal mice. Methods the contents of SOD,GSH--Px,MDA were measured in peripheral blood and liver cell. the number of CD4^ + T, CD8 ^+ T and IL-- 4 and IFN--γ of cyclosphamide --immunosuppressant mice were observed by the technique of ELISA and flow cytometry. Results Compared with Yupingfengsan apozem group and normal group, Yupingfengsan-- transform-- product group reduced the content of MDA (P 〈 0.01 ) and enhanced the activity of SOD,GSH--Px (P 〈 0.01). Yupingfengsan--transform-- product group improved the contents of CD4^+ T, CD8^ +T and IFN--γ than CP group (P 〈 0. 01 ,P 〈 0. 05). The percentage of IL--4 had no difference comparing with CP group. The ratio of CD4^+ T, CD8^+T in CP group was lower than Y Yupingfengsan--transform-- product group. Yupingfengsan--transform-- product group could improve the content of CD4^ + T, CD8 ^+T and IFN--γ than Yupingfengsan apozem group (P 〈 0.01). The percentage of IL--4,CD8 ^+T had no differenee comparing with Yupingfengsan apozem group. The ratio of CD4^ + T,CD8 ^+T in Yupingfengsan apozem group was lower than Yupingfengsan transformed products group. Conclusions Yupingfengsan-- transform--product increases the effects on antioxidant of normal mice and enhances the immune function of immunosuppressant mice.
