个体化裸鼠荷人肺癌动物模型的基因检测

摘    要:目的本研究是在建立具有个体化特征的肺癌移植瘤动物模型的基础上,对其肿瘤组织进行基因检测,为今后进行有关肺癌个体化治疗的一系列研究提供一个有效的实验工具。方法术后6小时内,将人非小细胞肺癌新鲜肿瘤组织,用接种针移植于BALB/C裸鼠,作为第一代移植模型,取第一代移植模型的肿瘤组织块,重复上述步骤,直至获得第三代移植模型。用定量寡聚核苷酸探针法检测部分第三代移植模型的肿瘤组织中EGFR DNA突变和RRM1 mRNA、TUBB3 mRNA表达情况。结果对3例动物模型肿瘤组织的基因检测显示,其EGFR DNA突变和RRMl mRNA、TUBB3 mRNA表达情况并不相同。2例分别有EGFR 18和EGFR 21外显子发生突变,1例无EGFR DNA突变。1例RRM1 mRNA呈高表达状态,其余2例RRM1 mRNA呈低表达状态。3例TUBB3 mRNA均呈低表达状态。结论个体化裸鼠荷人肺癌动物模型的建立方法简单可靠,重复性高,其肿瘤组织的基因突变和表达状况也各不相同。个体化动物模型的建立,为评估不同治疗方法对不同非小细胞肺癌患者的治疗效果,以及为今后开展的肺癌个体化治疗的一系列研究提供了一种全新、有效的评价和研究工具。

Gene test of individual human lung cancer model by xenotransplant tumor mass in nude mouse

Abstract:Objective To record gene test result of tumor, which is based on lung cancer xenotransplanted animal models with individual characteristic, to provide an effective experimental method for further individualized therapy study. Methods BALB/C nude mousewere injected with fresh non - small cell lung cancer tumor which was derived from patient within 6 hours after surgery. This is the first generation. Repeat this method to gain the third generation. Test the tumor in the third generation with PCR - ASO, to observe mutation of EGFR DNA and expression of RRM1 mRNA and TUBB3 mRNA. Results According to gene testresult of 3 animal tumor models, the mutation of EGFR DNA, the expression of RRM1 mRNA and TUBB3 mRNA were different. 2 models respectively showed mutation of EGFR 18 and EGFR 21 expressed region; no EGFR DNA mutation was found on the other model. One model~ RRM1 mRNA was on high expression of state, the other two models "were on low expression. All 3 models " TUBB3 mRNA showed low expression of state. Conclusions The method of xenotransplanting individual characterized human lung cancer tumor to immune deficient mice is easy, reliable and repeatable. The gene mutation and expression of tumor is different as well. The establishment of individual characterized animal model provides a new effective method to study and judge therapies of non - small cell lung cancer, and the further research on individual characterized therapies of lung
