摘    要:目的 构建7-OH香豆素-MnO2复合体系的荧光平台检测谷胱甘肽并用于药物质量控制. 方法 化学法合成MnO2纳米薄片并用于吸附7-OH香豆素形成复合体系,由于MnO2纳米薄片的内滤效应使得7-OH香豆素的荧光猝灭.谷胱甘肽(GSH)可与MnO2发生氧化还原反应,MnO2转变为可溶性的Mn2+从而释放出7-OH香豆素,恢复荧光.对比GSH加入前后荧光的恢复程度,荧光信号增大的幅度与GSH浓度正相关,可实现对GSH的高灵敏检测. 结果 该荧光平台可选择性识别GSH.在优化的实验条件下,荧光恢复值与GSH在0.03~400 μmol/L呈线性关系,检测限为0.01 μmol/L. 结论 构建的7-OH香豆素-MnO2复合体系的荧光平台可简便、准确地检测GSH,可用于实际药品中GSH的测定.

Abstract:Objective To construct a fluorescence platform based on 7-Hydroxycoumarin-MnO2 composites for the detection of glutathione and for its application in drug quality control.Method MnO2 nanosheets prepared by chemical method were used for the adsorption of 7-OH to form a composite system.Due to the internal filtering effect of MnO2 nanosheets,the fluorescence of 7-Hydroxycoumarin was quenched.The introduction of glutathione (GSH) reacted with MnO2 through redox reaction,and MnO2 was transformed into soluble Mn2+ to release absorbed 7-Hydroxycoumarin therefore recovered the quenched fluorescence.Compared the change in fluorescence intensity before and after the addition of GSH,the magnitude of the increase in fluorescence signal was positively correlated with the GSH concentration,therefore the high sensitive detection of GSH could be realized.Result The fluorescence platform can selectively recognize GSH.Under the optimized experimental conditions,the fluorescence recovery value was correlated with the GSH in the range of 0.03 μmol/L to 400 μmol/L in a linear relationship,and the detection limit was 0.01 μmol/L.Conclusion The construction of 7-HydroxycoumarinMnO2 composites can be used to detect GSH easily and accurately in medicinal testing.
