灵芝三萜组分GLA体内外抗肿瘤作用的研究

摘    要:目的评价灵芝三萜组分GLA的体内外抗肿瘤活性。方法采用四甲基偶氮唑蓝(MTT)法体外检测GLA对多种肿瘤细胞株的生长抑制作用;建立小鼠H22肝癌移植瘤模型,小鼠S180肉瘤移植瘤模型和人结肠癌Colon26裸小鼠移植瘤模型,评价GLA的体内抗肿瘤活性。结果 GLA对人肝癌细胞株SMMC-7721、人早幼粒白血病细胞株HL60和人Burkitt淋巴瘤细胞株CA46有较强抑制作用,IC50分别为为0.50,0.52,0.53 mg/mL,对人肝癌细胞株HepG2、人结肠癌细胞株SW480、人慢性粒细胞白血病细胞株K562和人胃癌细胞株SGC-7901也有一定的抑制作用,IC50分别为0.70,0.82,0.85 mg/mL和0.97 mg/mL,对人肺腺癌细胞株SP-CA-1和人恶性神经母细胞瘤株SH-SY5Y基本无抑制作用;体内实验表明,GLA在高剂量2 g/kg对小鼠H22肝癌细胞、小鼠S180肉瘤细胞及裸小鼠Colon26结肠癌细胞生长有一定抑制作用,其抑瘤率分别为65.6%,50.1%和46.5%。结论体内外试验均表明灵芝三萜组分GLA有较强的抗肿瘤作用。

Abstract:Objective To evaluate the anti-tumor effect of triterpene GLA from Ganoderma lucidum in vivo and in vitro.Methods Cell proliferation was assayed by methyl thiazolyl tetrazolium method to explore the cytotoxicity of GLA from Ganoderma lucidum in vitro.To investigate the anti-tumor effect in vivo,H22 Hepatoma and S180 sarcoma was induced in mice,and colon26 tumor was induced in nude mice.Results GLA showed inhibitory effects on the growth of tumor cell lines.The IC50 values to SMMC7221,HL60 and CA46 were 0.50...
