成年牛晶状体上皮细胞培养及超微结构的观察

目的 建立成年牛眼昌状体上皮细胞的培养方法，研究其在不同培养液中的生长特性并观察其超微结构。方法 组织块静置贴壁培养，应用细胞计数法和噻唑蓝（MTT）法检测细胞的生长情况，透射电镜观察所培养细胞的超微结构。结果 晶状体上皮细胞在高糖和低糖的DMEM培养液中的原代及传代培养生长均良好，在低糖DMEM中能更快速贴壁，电镜观察可见细胞内微丝丰富，高铛达，溶酶体多少不等，粗面内质风丰富和线粒体少，结论 组织场合胸置培养是合适的晶状体上皮细胞体外培养方法，细胞生长状况与培养液中含糖量关系不大，其较强的贴壁能力可能与所含丰富的微丝有关。

The Culture of Adult Bovine Lens Epithelial Cells and its Ultrastructural Observation

Objective\ To establish an adult bovine lens epithelial cells culture system, and investigate the growth characteristics of the cells in different culture media and its ultrastructure.\ Methods\ Lens epithelial cells were planted and cultured.\ The cell growth was examined by cell counting and MTT method.\ Its ultra structure was observed through transmission electron microscopy.\ Results\ Lens epithelial cells grew well during planting culture.\ Its adherence was faster in the DMEM medium containing low glucose(P>0.05).\ Rich microfilaments, well\|developed Golgi complex, many profiles of rough endoplasmic reticulum, various amounts of lysosome, and small amounts of mitochondria were found within the cells.\ Conclusion\ DMEM medium is recommended the planting culture for lens epithelial cells.\ The growth of the cells was not related to glucose amounts in the medium.\ The adherence capability of cells might coincide with its rich microfilaments.\ This system will play a fundamental role in further biological characteristics and pharmacotherapeutic research of lens epithelial cell.
