摘    要:研究不同产地及生长方式的金线莲样本间遗传多样性和亲缘关系。 方法 以20份福建金线莲新鲜叶片为实验材料,采用ISSR-PCR分子标记技术,筛选出15条多态性好、扩增条带清晰的ISSR引物。经琼脂糖凝胶电泳成像后,统计其扩增条带数,运用NTSYS-Pc 2.1和POPGENE 32软件进行UPGMA聚类分析。 结果 共扩增出130条清晰可见的条带,多态性比率达100%,遗传相似系数分布在0.647 6~0.971 4,遗传距离分布在0.029 0~0.434 5,其中编号为3和4的样本具有较近的亲缘关系。聚类分析结果显示,当相似系数取0.78时,20份样本被分为3类,可以区分不同的产地和种类。 结论 运用ISSR标记技术,可以从分子水平上揭示福建省内金线莲的遗传多样性。

Abstract:Objective To study the genetic diversity and genetic relationship among different habitats and growth patterns of Anoectochilus roxburghii. Methods A total of 20 fresh leaves of Fujian Anoectochilus roxburghii were used as samples, and 15 ISSR primers with good polymorphism and amplification were screened out by ISSR-PCR molecular marker technology. After the agarose gel electrophoresis, the number of amplified bands was counted. The data were analyzed by NTSYS-Pc 2.1 and POPGENE 32 software, and clustering were performed by UPGMA method. Results A total of 130 clearly visible bands were amplified, and the polymorphism ratio is up to 100%. The genetic similarity coefficient ranged from 0.647 6 to 0.971 4, and the genetic distance distribution was 0.029 0 to 0.434 5. The genetic relationship between samples No.3 and No.4 was the closest. The clustering results showed that when the similarity coefficient is 0.78, 20 samples were divided into three categories, which could distinguish between different habitats and varieties. Conclusion The ISSR marker technology could reveal the genetic diversity of Anoectochilus roxburghii at the molecular level.
