全脑照射对小鼠室管膜下区GFAP表达及细胞增殖的影响

摘    要:目的:探讨60Co-γ射线全脑照射对小鼠室管膜下区星形胶质细胞GFAP表达及细胞增殖的影响。方法:将56只昆明小鼠随机分成4组,每组14只,分别以0,5,15 Gy和30 Gy剂量进行照射,分别观察照射后1周和4周室管膜下区的GFAP和BrdU阳性细胞形态和数目。结果:①照射后1周,15 Gy组和30 Gy组室管膜下区GFAP阳性细胞数明显增多(P〈0.01),BrdU阳性细胞数明显减少(P〈0.01);②照射后4周,15 Gy组室管膜下区GFAP阳性细胞数仍明显增多(P〈0.01),但其BrdU阳性细胞数恢复到对照组水平;30 Gy组室管膜下区GFAP阳性细胞数恢复到对照组水平,但BrdU阳性细胞数仍明显减少(P〈0.01)。结论:放射后室管膜下区GFAP表达可能与神经干细胞的增殖有关。

Abstract:Objective:To investigate the astrocytes;glial fibrillary acidic protein(GFAP) expression of astrocytes in subventricular zone(SVZ) of mice and the effects of GFAP-positive cells on proliferation in SVZ after 60↑Co-γRay whole brain irradiation.Methods:Fifty-six mice were randomly divided into control,5 Gy,15 Gy and 30 Gy groups,14 mice each group.The reactive changes of the BrdU-positive cells and GFAP-positive cells in SVZ were observed in one week and four weeks respectively for irradiation.Results:①The GFAP-positive cells in both 15 Gy and 30 Gy groups were increased significantly(P〈0.01),while the BrdU-positive cells were decreased significantly(P〈0.01) in one week.② In four weeks,the GFAP-positive cells in SVZ of 15 Gy group were still increased significantly(P〈0.01),but the BrdU-positive cells increased and had no difference with the control.The GFAP-positive cells in SVZ of 30 Gy group decreased and had no difference with the control,but BrdU-positive cells were still deceased decreased significantly(P〈0.01).Conclusion:There is correlation between the expression change of GFAP and proliferation of neural stem cells in SVZ.
