猪、熊胆粉主要成分对ECV304细胞缺氧损伤保护作用的比较

摘    要:目的 比较猪胆粉与熊胆粉中胆酸类主要成分对细胞缺氧损伤保护作用的差别 ,为猪胆粉替代熊胆粉用于中风病治疗提供实验依据。方法 培养的ECV30 4细胞 ,用连二亚硫酸钠造成缺氧损伤 ,并分别以MTT比色法、LDH漏出率和台盼蓝染色法进行检测 .比较猪胆粉与熊胆粉中胆酸类主要成分对细胞缺氧损伤保护作用的差别。结果 应用连二亚硫酸钠造成ECV30 4细胞明显损伤 ,而猪去氧胆酸、牛磺熊去氧胆酸和牛磺鹅去氧胆酸钠均使ECV30 4细胞的缺氧损伤明显减轻 ,与对照组间有显著差异 ,但三组间没有明显差别。结论 猪去氧胆酸对ECV30 4细胞缺氧损伤具有与牛磺熊去氧胆酸相类似的保护效应 ,提示猪胆粉可替代价格昂贵的熊胆粉用于中风病的治疗。

Abstract:Objectives To compare the protective effects against the injury by cytohypoxia of the main cholic acid like component of porcine gall powder with those of bear gall powder, in order to provide experimental data for the possible substitution of porcine gall powder for bear gall powder for treating apoplectic syndrome in TCM clinical practice. Methods The injury by cytohypoxia was induced by sodium dithionite in the cultured cells of ECV304 cell line; MTT, LDH leakage rate test, and trypanblau staining were used to find the difference between the protective effects against the injury by cytohypoxia of hyodeoxycholic acid (HDCA), the main component of porcine gall powder, and the same effects of tauroursodesoxycholic acid (TUDCA) and taurochenodesoxycholic acid (TCDCA), the main components of bear gall powder. Results Sodium dithionite could induce remarkable injury in the cells of ECV304 cell line; all HDCA, TUDCA and TCDCA could relieve the injury by cytohypoxia induced by sodium dithionite significantly as compared with a control ( P <0 001) while no remarkable difference between the effects of HDCA, TUDCA and TCDCA was found. Conclusions HDCA possesses an effect against the injury by cytohypoxia of the cells of ECV304 cell line, which is similar to that of TUDCA, which suggests that porcine gall power can be used to substitute bear gall powder for treating apoplectic syndrome.
