通脑精胶囊对IR大鼠局灶性脑缺血血浆t-PA及PAI-I和DD的影响

摘    要:高糖饲料喂养复制胰岛素抵抗(IR)大鼠模型,采用电凝大脑中动脉造成局灶性脑缺血.结果:与空白对照组及模型组比较,IR基础上的局灶性脑缺血大鼠血清胰岛素(Ins)水平明显升高(P<0.01),胰岛素敏感指数(ISI)显著降低(P<0.01),血浆组织型纤溶酶原激活物(t-PA)含量及活性显著降低(P<0.01),组织型纤溶酶原抑制物(PAI-1)和D-二聚体(DD)含量明显升高(P<0.01);经通脑精胶囊治疗后,上述指标均有明显好转(P<0.01).提示IR大鼠局灶性脑缺血后存在明显的IR状态及纤溶活性的降低,而通脑精胶囊具有逆转IR及增强纤溶活性作用,推测这可能是该方治疗急性脑梗死的作用机制之一.

Abstract:The rat model of insulin resistance was established by the method of feeding high carbohydrate forage, and then, the middle cerebral artery electrocoagulation was performed on the insulin resistant model. The effects of Tongnaojing Capsules (TC) on the level of serum insulin, insulin sensitivity index (ISI), the level and activity of plasmatic tissue plasminogen activator (t PA), and the levels of plasminogen activator inhibitor 1 (PAI 1) and D dimer (DD) were observed in the rat model of insulin resistance under cerebral ischemia. The results showed that compared with those in the rats in the blank control and the model group, the level of serum insulin markedly increased ( P <0 01), ISI significantly decreased ( P <0 01), the level and activity of plasmatic t PA markedly reduced ( P <0 01), and the levels of PAI 1 and DD significantly increased ( P <0 01) in the insulin resistant rats under cerebral ischemia; after the treatment with TC, the abnormality of these criteria was remarkably reduced ( P <0 01), which suggests that the insulin resistant rats under cerebral ischemia are in an obvious state of insulin resistance and fibrinolytic activity decrease, and TC can reverse their insulin resistance and enhance their fibrinolytic activity, which might be one of the mechanisms of TC in curing acute cerebral infarction.
