束缚应激大鼠AMPA受体和相关蛋白变化及逍遥散对其影响

摘    要:目的观察慢性束缚应激状态下海马及杏仁体AMPA受体和相关蛋白的变化及逍遥散的作用。方法使用捆绑的方法制作慢性束缚应激动物模型,并用逍遥散进行干预,分别于7 d后和21d后检测模型大鼠海马CA1区、基底外侧杏仁核(BLA)谷氨酸受体2/3(GluR2/3)、N-乙基顺丁烯二酰亚胺敏感性的融合蛋白(NSF)、PKC作用蛋白1(PICK1)的积分吸光度(IOD)和阳性神经元数。结果7 d模型组大鼠海马CA1区和BLAGluR2/3 IOD较7 d正常组显著增高(P<0.01,P<0.05);7 d模型组海马CA1区GluR2/3阳性神经元数及PICK1 IOD较7 d正常组增高(P<0.05,P<0.01),7 d治疗组(逍遥散)上述指标与7 d模型组相比有显著差异(P<0.01,P<0.05),与7 d正常组比较无差异;21 d模型组与21 d正常组比较,海马CA1区GluR2/3 IOD显著降低(P<0.05)、NSF IOD升高(P=0.06),BLA GluR2/3阳性神经元数增多(P=0.090)、PICK1阳性神经元数减少(P=0.075),21 d治疗组上述指标与正常组比较无显著性差异。结论7 d和21 d束缚应激对AMPA受体影响的变化趋势不同,海马CA1区与BLA的变化趋势也不一致,逍遥散对21 d束缚应激状态下AMPA受体的变化调节较强。

Changes of AMPA receptors and related protein in immobilization stress rats and effect of Xiaoyao Powder

Abstract:Objective To observe changes of AMPA receptor and related protein in the hippocampus and amygdala in rats with chronic immobilization stress and Xiaoyao Powder's effect.Method The immobilization stress model rat was established by tied 3hours per day and treated with Xiaoyao Powder.They were assigned into 7days model group,21days model group,7days treated group and 21days treated group.Besides,7days normal group and 21days normal group were set for control.After 7 days and 21 days,the integral optical density(IOD) of glutamate receptor2/3(GluR2/3),N-ethylmaleimide sensitive factor(NSF) and protein interacting with C-kinase 1(PICK1) as well as the number of positive neuron in the rat hippocampal CA1 region and basolateral amygdala(BLA) were detected by immunohistochemical stain and computerized graphic analysis techniques.Resluts The IOD of GluR2/3 in hippocampal CA1 region and BLA in 7days model group were significantly higher than that of 7days normal group(P<0.01,P<0.05);The positive neurons of GluR2/3 and the IOD of PICK1 in hippocampal CA1 region in 7days model group were higher than that of 7days normal group(P<0.05,P<0.01),while in 7days treatment group(Xiaoyao Powder) those indexes were significantly different from those in 7days model group(P<0.01,P<0.05),but no significant difference was showed when compared with 7days normal group.Compared with 21days normal group,the IOD of GluR2/3 increased significantly(P<0.05),NSF increased(P=0.060) in hippocampal CA1 region,and the positive neurons of GluR2/3 increased(P=0.090),PICK1 in BLA decreased(P=0.075) in 21days model group,while no significant difference of those items in 21days treatment group was found when compared with 21days normal group.Conclusion There were different trends of AMPA receptor changes in 7 days and 21 days immobilization stress rats,and in hippocampal CA1 region and BLA.The regulative effect of Xiaoyao Powder on AMPA receptors in 21 days immobilization stress rats was more extensive than in 7 days rats.
