醒脾开郁方对抑郁症大鼠模型的中枢单胺类递质的影响

摘    要:目的研究醒脾开郁方对抑郁症大鼠模型海马及皮层的单胺类递质的影响.方法结合分养及慢性不可预见性应激的方法造成大鼠抑郁模型,应用高效液相色谱-电化学检测器观察大鼠海马及皮层单胺类递质和其代谢产物的变化,以探讨醒脾开郁方抗抑郁的机制.结果模型组大鼠海马及皮层5-羟色胺(5-HT)含量明显低于正常组,海马3,4二羟基苯乙酸(DOPAC)、高香草酸(HVA) 、DOPAC/多巴胺(DA)、5-羟吲哚乙酸(5-HIAA)/5-羟色胺(5-HT)及皮层5-HIAA/5-HT均明显高于正常组.醒脾开郁方可明显升高慢性应激抑郁模型大鼠海马及皮层5-HT,降低海马DOPAC、HVA含量,并降低海马 DOPAC/DA 、5-HIAA/5-HT和皮层5-HIAA/5-HT.结论提示醒脾开郁方通过降低海马和皮层5-HT及DA的代谢,相对升高海马5-HT、DA及皮层5-HT含量而发挥抗抑郁作用.

Abstract:Objective To investigate the effect of Xingpikaiyu Formula(XF)on monoamine neurotransmitters in cortex and hippocampus of rat model of depression.Method The rat model of depression was established by using the methods of separation and chronic unpredictable stress.The changes of monoamine neurotransmitters and their metabolites in cortex and hippocampus were determined by HPLC_electrochemistry.Results Compared with the normal group,5_HT in cortex and hippocampus of the model group was obviously decreased.While the levels of DOPAC,HVA,DOPAC/DA,5_HIAA/5_HT of hippocampus and 5_HIAA/5_HT of cortex in the model group were higher significantly than that of the normal group.XF obviously increased 5_HT in cortex and hippocampus and decreased DOPAC and HVA in hippocampus.XF also decreased DOPAC/DA,5_HIAA/5_HT of hippocampus and 5_HIAA/5_HT of cortex.Conclusion XF has an anti_depression effect through inhibiting the metabolism of 5_HT and DA in cortex and hippocampus,and relatively increases the levels of 5_HT and DA in hippocampus and 5_HT in cortex.
