黄芩、栀子配伍对大鼠局灶性脑缺血再灌注模型缺血级联反应的影响

摘    要:通过对黄芩、栀子配伍阻抑脑缺血级联反应的作用特征分析,探讨两者配伍治疗缺血性中风的作用环节及作用部位.用线栓法建立大鼠大脑中动脉缺血再灌注模型,观察黄芩、栀子及两者配伍对大鼠神经症状、脑组织SOD、丙二醛(MDA)、一氧化氮合成酶(NOS)、Na+-K+-ATP酶及血浆凝血因子Ⅷ相关抗原(vWF)等的影响.结果显示,黄芩、栀子及其配伍均能改善大鼠神经功能缺损症状;黄芩、栀子单用对脑组织SOD、MDA和血浆vWF均未见明显影响,而两者配伍则可显著提高脑组织SOD活性,降低脑组织MDA和血浆vWF含量,但对于NOS及 Na+-K+-ATP酶的活性,则有减效、减毒趋势.实验证明,中药组分配伍后的作用不是单纯的效应累加,而是以效应环节的生物特性表现为协同增效或减毒等复杂的现象.

Abstract:The mechanism of huangqin (the dried root of Scutellaria baicalensis) and zhizi (the dried fruit of Gardenia jasminoides) used in combination for treating ischemic apoplexy was investigated by analyzing the characteristics of the effects of huangqin and zhizi used in combination in inhibiting the cerebral ischemic cascade reaction. The rat model of ischemia and reperfusion of middle cerebral artery was established by the thread occluding method; and the effects of huangqi and zhizi used in combination on neural symptoms, the levels of SOD, MDA, NOS and Na + K + ATPase in the brain and the level of vWF in the plasma were observed. The results showed that both the single use of huangqin or zhizi and the combinative use of huangqin and zhizi could improve the symptoms of neural function defect; the single use of huangqin or zhizi had no obvious effects on SOD, MDA and vWF, while the combinative use of huangqin and zhizi could enhance the activity of SOD and lower the levels of MDA and vWF, at the same time, showing an effect diminishing and attenuating tendency toward the activities of NOS and Na + K + ATPase. The results suggest that the effects of the combinative use of huangqin and zhizi are not the simple addition of their respective effects but some complicated biological phenomena.
