何首乌药材HPLC指纹图谱研究

摘    要:目的 建立何首乌药材反相高效液相色谱(RP-HPLC)指纹图谱分析方法,为有效控制何首乌药材的质量奠定基础.方法 采用RP-HPLC法,色谱柱为SunfireTM C18,乙腈-0.1%甲酸水梯度洗脱,检测波长为280 nm,流速为1.0 mL/min,分析时间为60 min,分析了10批生首乌药材和10批制首乌药材的HPLC指纹图谱.结果 在选定的色谱条件下通过相似度分析确定13个色谱峰构成生首乌药材指纹图谱的特征峰,11个色谱峰构成制首乌药材指纹图谱的特征峰,可用于不同产地何首乌药材的指纹图谱的测定和质量评价.结论 采用RP-HPLC方法所建立的指纹图谱具有稳定、重复性好的特点,不但可以用于何首乌药材质量控制,还可以作为探索中药何首乌有效物质基础研究的有效手段.

Abstract:Objective To set up a analysis method of fingerprint chromatogram of Polygonum multiflorum Thunb.by RP-HPLC,and lay a basis for the quality control of medical material of Polygonum multiflorum Thunb.Method RP-HPLC was used on a chromatographic column of SunfireTM C18.Acetonitrile-formic acid water solution(0.1%) was taken as the gradient elution,and the mobile phase was at flow rate of 1.0 mL/min.The detection wavelength was at 280 nm and the analysis time was 60 min.HPLC fingerprint chromatograms of 10 batches of crude material of Polygonum multiflorum Thunb.and 10 batches of prepared Polygonum multiflorum Thunb.were analyzed.Result Under the above chromatographic conditions the characteristic peak of fingerprint chromatograms of crude material of Polygonum multiflorum Thunb.was composed of 13 chromatographic peaks,and that of prepared Polygonum multiflorum Thunb.was composed of 11 chromatographic peaks through the analysis of similarity.The results could be used for the determination and quality appraisal of the fingerprint chromatogram of Polygonum multiflorum Thunb.produced from different areas.Conclusion The fingerprint chromatogram set up by RP-HPLC is stable and repeatable,which can be used for not only the quality control but also the research on effective principles of Polygonum multiflorum Thunb.
