摘    要:目的用一株鹅源H5N1亚型禽流感病毒感染小鼠,观察其对小鼠的致病性及在小鼠间的传染性。方法将小鼠分为正常组、攻毒组、同居组,攻毒组乙醚麻醉后滴鼻感染AIV,观察各组食欲、反应、呼吸及有无皱毛、弓背、死亡等,测量体温及体重;检测相应抗体水平、小鼠排毒情况及小鼠各器官带毒情况,并观察病理组织学变化。结果攻毒组出现食欲反应下降,呼吸急促,皱毛,死亡;体重、体温下降。同居组未出现感染。攻毒组攻毒后12 h至第9天可从肺组织分离出病毒,第10天始出现相应抗体;感染小鼠的肺、心肌、肝、脾、肾和脑组织都有不同程度的病理改变。结论 AIV对小鼠具有致病性,AIV能跨越种系障碍直接感染哺乳动物, 但在小鼠间无传染性。

Abstract:Objective A Goose-origin H5N1 avian influenza virus was analyzed for its pathogencity in mice and the fact whether this disease was infectious among mice. Methods Anesthetized mice were inoculated intranasally with influenza virus isolates. Throughout the 14-day experiment clinical signs were observed; body weight and body temperature were recorded; the blood samples were collected for analyzing the anti-H5 antibodies; tissue samples from major organs such as lungs, brains, and kidneys were processed for virus isolation and histopathological analysis. Results The H5N1 AIV caused mild to severe diseases in mice, characterized by clinical signs, such as hypothermia, weight loss and death.Virus could be isolated from the lungs of infected mice from 12h.p.i to day 9.p.i.Anti-H5 antibodies were detected and variable degrees of lesions were observed in lungs, brains and other organs. The group of mice living with the infected mice were not infected. Conclusion The H5N1 viruses can cross the species barrier and cause infection in mice without a prior adaptation. However, this disease was not infectious among mice.
