摘 要: | 目的探讨单核细胞趋化因子-1(MCP-1)在瑞舒伐他汀钙干预大鼠肺动脉高压中的作用。方法将24只Wistar雄性大鼠分为空白对照组(C组)、野百合碱诱导肺动脉高压组(M组)和瑞舒伐他汀钙组(R组),M组和R组给予野百合碱皮下注射,R组同时给予10 mg/(kg·d)瑞舒伐他汀钙灌胃,6周后检测各组大鼠平均肺动脉压力(mPAP)、右心室/(左心室+室间隔)重量比RV/(LV+S)、肺组织单核细胞趋化因子-1(MCP-1)mRNA和蛋白表达。结果野百合碱可以上调MCP-1表达,诱导mPAP升高,提高RV/(LV+S),瑞舒伐他汀钙可以下调MCP-1表达,降低mPAP,降低RV/(LV+S)。结论瑞舒伐他汀钙可能通过下调MCP-1改善大鼠肺动脉高压。

Abstract: | Objective To investigate the effect of rosuvastatin calcium on rat pulmonary arterial hypertension induced by monocrotaline. Methods 24 male Wistar rats were equally divided into control group (C group), monoerotaline-induced pulmonary arterial hypertension group(M group)and rosuvastatin group(R group). M group and R group received monocrotaline subcutaneous injection, and R group also received 10 mg/kg rosuvastatin by gavage daily for six weeks after injection. After six weeks, mean pulmonary arterial pressure(mPAP), right ventricular hypertrophy index [RV/(LV+S)] ,MCP-1 mRNA and protein expression in lung were detected. Result Monocrotaline up regulated MCP-1,elevated mPAP and RV/(LV +S), whereas rosuvastatin down regulated MCP-1,lowered mPAP and RV/(LV+S). Conclusion Rosuvastatin attenuates rat pulmonary arterial hypertension via down regulation of MCP-1. |
