芩荑合剂对哮喘大鼠STAT6表达的影响

摘    要:目的:观察芩荑合剂对哮喘大鼠肺组织模型信号转导子和转录激活子(STAT6)蛋白含量及其mRNA表达影响,探讨芩荑合剂对支气管哮喘的作用机理。方法:用卵清蛋白建立大鼠哮喘模型,将48只SD大鼠随机分为正常对照组、哮喘模型组、地塞米松组、芩荑合剂低剂量组、芩荑合剂中剂量组、芩荑合剂高剂量组,其中正常对照组、哮喘模型组给予生理盐水灌胃,芩荑合剂高、中、低剂量分别按生药量5.4、2.70、1.35 g/(kg·d)灌胃,地塞米松按1.4 mg/(kg·d)灌胃,采集肺组织标本,分别采用实时荧光定量PCR(Realtime PCR)法和免疫组化法检测哮喘大鼠模型STAT6m RNA及其蛋白表达。结果:中、高剂量组STAT6 mRNA及蛋白表达较哮喘组显著减弱,有显著差异(P〈0.01),与地塞米松组比较无统计学意义。结论:芩荑合剂通过降低哮喘大鼠STAT6蛋白含量以及减弱STAT6 mRNA在气道上皮细胞的表达,来干预气道炎症,从而达到治疗支气管哮喘的目的。

Abstract:Objective : To investigate the effect of Oinyi Mixture on the amounts of STAT6 mRNA and protein in lung in asthmatic rats, and to determine the mechanism of Qinyi Mixture on bronchial asthma. Methods : The rat asthma model was made by using ovalbumin. 48 SD rats were randomly divided into normal control group, asthma model group, dexamethasone group, Qinyi mixture low--, medium-, high-dose groups. Normal control group, asthma model group were administrated saline lavage, Qinyi mixture high-, medium- and low-doses respectively were administrated Qinyi mixture 5.4 g/kg,2.70 g/kg, 1.35g/ kg., dexamethasone 1.4 mg/kg every day. Lung tissue was collected, and STAT6 mRNA and protein were detected with Reahime PCR assay and immunohistochemical assay respestively. Results : The expressions of STAT6 mRNA and protein in medium and high dose groups of Qinyi Mixture were reduced significantly compared with that of model group (P〈0.01), also there was no statistics difference compared with dexamethasone group. Conclusions : Qinyi Mixture can decrease the amounts of eosinophil in serum and BALF and the expressions of STAT6 mRNA and protein in lung in asthmatic rats, though which to intervene airway inflammation so as to achieve the purpose of treating bronchial asthma.
