K~+对三链DNA形成的影响

摘    要:用电泳迁移分析方法及DNaseⅠ足迹实验研究了脱氧寡核苷酸与乙肝病毒(HBV)核衣壳启动子(Cp)片段形成三链DNA中的K~+效应。K~+对三链DNA形成的抑制作用大小取决于不同的脱氧寡核苷酸。碱基的错配可使K~+的抑制作用显著增强,提示K~+的效应强弱可能与三链结构中碱基准积的连续程度有关。因此反应体系中K~+的存在能提高三链DNA形成的特异性。

Abstract:The effects of K+ on the triplex-formation between oligodeoxyribonucleotides and core promoter(Cp) fragment of hepatitis B virus (HBV) were investigated by electrophoretic mobility shift analysis andDNase I footprinting. The inhibitory effects of K+ were determined by different oligodeoxyribontlcleotides.Base mismatching could enhance the inhibitory effects of K+,suggesting that the effects of K+,were inter-related with the succession of base stacking in triplex DNA. In conclusion,the existence of K+ in reactionbuffer may increase the speciflcity of triplex-formation.
