摘    要:目的 对链霉菌胞外多糖、灵芝多糖和米糠多糖体外抑制HIV gp41六股螺旋束核心结构的形成进行比较研究.方法 采用作用于HIV gp41的HIV融合抑制剂的高通量筛选方法(夹心ELISA法)检测3种多糖对HIV gp41六股螺旋束核心结构形成的抑制作用.结果 和结论链霉菌胞外能有效抑制HIV胞膜糖蛋白gp41六股α-螺旋束核心结构的形成,其半效抑制浓度(IC50)为(145.48±7.25)mg/L-1,而灵芝多糖和米糠多糖不具有这种抑制活性.

Abstract:OBJECTIVE: To compare the in vitro inhibitory effect of expolysaccharides from Streptomyces, polysaccharides of Ganoderma lucidum and rice bran on six-alpha-helix bundle formation of HIV gp41 protein. METHODS: The amount of six-alpha-helix bundle formed in the presence of N36 and C34 was tested by ELISA in response to treatments with different doses of polysaccharides. RESULTS: Expolysaccharides from Streptomyces potentially inhibited six-alpha-helix bundle formation with the effective concentration (IC(50)) of 145.48-/+7.25 mg /L. Polysaccharides of Ganoderma lucidum and rice bran showed no effect on the six-alpha-helix bundle formation. CONCLUSION: Expolysaccharides from Streptomyces can inhibit the six-alpha-helix bundle formation of HIV gp41, whereas polysaccharides of Ganoderma lucidum and rice bran do not exhibit such activity.
