摘    要:摘要:目的探究血清miR-103在乳腺浸润性癌患者中表达的诊断价值及其与乳腺癌临床病理特征的关系。方法搜集50例首
治的乳腺癌患者的血清及相应的石蜡包埋组织和50例正常女性的血清作为对照,从血清中提取总RNA,运用荧光定量PCR的
技术检测miR-103在乳腺癌和正常人血清中的表达情况。运用SPSS16.0软件行t检验、ANOVA、χ2检验、秩和检验及MedCalc
软件进行受试者工作特征曲线(ROC)分析。结果与对照组相比,miR-103在乳腺癌患者血清中表达量显著上调(P<0.01),血清
中miR-103 上调的相对表达量与乳腺癌临床分期(P<0.05)、淋巴结转移(P<0.05)相关。miR-103 所得ROC 曲线下面积为
84.3%,区别乳腺癌和正常人的敏感性和特异性分别为84%、70%。结论血清miR-103可作为乳腺癌一项潜在的诊断标记物,
并对乳腺癌的临床病理特征具有提示作用。

Abstract:Objective To evaluate the diagnostic value of serum miR-103 for breast cancer and its correlation to the clinicopathological features of the patients.Methods We collected the serum samples and corresponding formalin-fixed paraffin-embedded(FFPE) surgical specimens from 50 breast cancer patients,using the serum samples from 50 healthy women as the control.The total RNA was extracted from the samples for quantitative analysis of miR-103 using real-time RT-PCR.Results The serum levels of miR-103 expression were significantly higher in the cancer patients than in the healthy control group(P<0.01).In the cancer patients,high miR-103 expression was significantly correlated to advanced clinical stage(P<0.05) and lymph node metastasis(P<0.05).miR-103 showed a receiver operating characteristic(ROC) curve area of 84.3%,and a sensitivity of 84% and a specificity of 70% in discriminating breast cancer patients from the control subjects.Conclusion Serum miR-103 can serve as a potential diagnostic marker for breast cancer and provides useful information of the clinicopathological features of the patients.
