抗小鼠激活T淋巴母细胞抗体的诱导及鉴定

用伴刀豆蛋白A(Con A)体外诱导的T淋巴母细胞免疫同系小鼠获得的抗体,能与不同品系小鼠的T淋巴母细胞和杀伤性T淋巴细胞株(CTLL2)起结合反应,并能抑制CTLL2细胞的增殖,阻断Con A诱导小鼠脾细胞转化及混合淋巴细胞反应等生物学特性,结果提示:该抗体具有抗小鼠白细胞介素2受体的特征。

INDUCTION AND IDENTIFICATION OF AN ANTIBODY AGAINST THE SYNGENEIC CONCANAVALIN A ACTIVATED MOUSE SPLEEN CELL

An activated anti mouse T cell antibody (anti MuTac) was obtained from BALB/C or C57BL/6J mice immunized by intraperitoneal syngeneic concanavalin A (Con A) activated mouse spleen cells. Immunizing the C577BL/6J -mice with CTLL2 cells, we got an anti CTLL2 antibody.Analysis of the anti MuTac and anti OTLL2 antibody by indirect combined tests showed that both antibodies were bound to murine Oon A activated T lymphoblasts, IL2 dependent CTLL2 and Lacl cell lines,but not to resting thymocytes, splenocytes or mesenteric lymph node cells.As tested by biological assay, the anti MnTac antibody could (1) significantly inhibit proliferation of IL2 dependent CTLL2 cell line; (2) completely inhibit proliferation of spleen cells by Con A stimulation in vitro. The inhibitory effect was dose and phase dependent, i. e., the inhibitory rate was over 90% when the antibody was added the culture system at the early stage (0-6h). When it was added after 36 h, no inhibitory effect was seen; and (3) also considerably block the mouse MLR. It was shown that no MHO restriction was seen in all experiments.In conclusion, these results indicate that anti MuTac and/or anti CTLL2 antibodies may be considered to be an anti mouse IL2 receptor antibody.
