摘    要: 目的 建立简单、快速和灵敏的高效液相色谱串联质谱法以测定人血浆中拉莫三嗪的浓度。 方法 血浆样品经甲醇直接沉淀蛋白后,采用Thermo Hypurity C18柱(4.6×250 mm,5 μm)分离,流速为0.20 mL/min,以甲醇∶0.1%甲酸=80∶20为流动相等度洗脱。通过电喷雾离子源进入质谱,以二级质谱多反应监测(multiple reaction monitoring,MRM)方式进行检测。结果 拉莫三嗪的线性范围为8.19~2 096 ng/mL,平均相对回收率在80% ~ 120%范围内,日内和日间变异均 <15%。结论 本法简单、快速、重现性好,能够用于拉莫三嗪的人体药代动力学性研究。

Abstract:Objective To develop a simple,quick and sensitive method of liquid chromatography-electrospray tandem mass spectrometry to determine lamotrigine in human plasma. Methods After precipitation with methanol,plasma sample was separated on a Thermo Hypurity C18 column(4.6×250 mm,5 μm),with mobile phase methanol∶0.1% formate acid(80∶20) and flow rate 0.20 mL/min.The eletrospray tandem mass spectrum in the positive mode and multiple reaction monitor(MRM) were used to detect the concentration of lamotrigine. Results The response was linear over the range of 8.19-2 096 ng/mL for lamotrigine.The relative recovery of lamotrigine was in the range of 80%-120%.The interday and intraday relative standard deviations were less than 15%. Conclusions This method is simple,quick and reproducible for pharmacokinetics and bioequivalence research of lamotrigine.
