吗啉、2-巯基苯并噻唑及其两种衍生物在豚鼠上致敏力的比较

摘    要:本文研究吗啉(morpholine,M)、2-巯基苯并噻唑(2-mercaptobenzothiazole,MBT)及其两种衍生物4,4’-二硫双吗啉(4,4-dithiodimorpholine,DTDM)及吗啉代巯基苯并噻唑(morpholinylmercaptobenzothiazole,MMBT)在豚鼠上的致敏力,5%及10%DTDM、MMBT及MBT可致刺激反应,M则否。用5%浓度作诱导试验,2%、0.5%及0.1%作激发试验。DTDM及MMBT组激发试验均阳性,其强度前者明显大于后者。MBT组70%阳性。M组均阴性。交叉敏感试验中DTDM组60%对MMBT发生反应,30%对M反应;MMBT组80%对MBT及20%对M发生反应;MBT组30%对MMBT出现反应。致敏潜力依次为DTDM、MMBT及MBT。二硫键、一硫键及硫氢基在本组化学物致敏中似发挥一定作用。

Abstract:The aim of this study was to determine the sensitizing potency of morpholine (M), 4, 4'-dithiodimorpholine(DTDM), morpholinylmercaptobenzothiazole(MMBT) and 2-meroaptobenzothiazole (MBT) in guinea pigs. 5% and 10% DTDM, MMBT and MBT produced irritation reactions. M up to 10% failed to irritate. For all of the compounds studied, 5% in petrolatum was used as the induction concentration; 2%, 0.5% and 0.1% in petrolatum were used as challenge concentrations. All of the guinea pigs treated with DTDM and MMBT were sensitized; 70% of the animals were sensitized to MBT, none of the animals treated with M were sensitized. When the animals were subsequently retested for cross-sensitization, the following observation were made: 60% of the DTDM-sensitized animals reacted to challenge with MMBT, 30% of the animals sensitized to DTDM reacted to challenge with MBT, and 20% to challenge with M; 30% of the MBT-sensitized animals reacted to MMBT. The rank order of sensitization potential in guinea pigs as observed from this study is DTDM, MMBT and MBT. It appears that the disulfide bond, sulfur linkage and the sulfhy-aryl group may each play a role in the sensitizing capacity of these compounds.
