摘    要:目的通过观察杜仲总黄酮对新生大鼠颅盖骨体外培养成骨细胞增殖的影响,筛选杜仲防治骨质疏松的药效成分。方法乙醇分段法提取杜仲皮中总黄酮,以25μg/ml、50μg/ml、100μg/ml浓度分别加入新生Wistar大鼠颅骨分离培养的成骨细胞中,用MTT法评价对成骨细胞增殖的影响。结果50、100μg/ml杜仲总黄酮剂量组的OD570吸光值明显增高,与对照组比较,差异显著(P〈0.05)。结论杜仲总黄酮能直接促进体外成骨细胞的增殖。

Abstract:Objective Effective component to osteoporosis was screened by observing effect of total flavone of Eucommia bark on proliferation of the osteoblast obtained from novel bomed Wistar rat's cranium. Methods The total flavone were extracted from Eucommla bark by ethanol of differential concentrations (from 60%-90%), and the osteoblast was cultured in total flavone of differential concentrations which were diluted at 25 μg/ml, 50μg/ml and 100 μg/ml in vitro. The effects on proliferation of the osteoblast were evaluated by MTT. Results By control, the cells cultured with the total flavone at 50 and 100 μg/ml had higher optical absorptione at 570 nm and it meant the total flavone had notable effect on the proliferation of the osteoblast at these two concentrations. Conclusions The total flavone of Eucommia bark can improve proliferation of osteoblast in vitro(P〈0.05).
