高压氧对化疗药物体外诱导急性髓性白血病细胞Bax和Bcl—2基因表达的影响

摘    要:①目的 采用体外实验方法探讨高压氧(HBO)对化疗药物体外诱导急性髓性白血病(AML)细胞Bax和bcl-2基因表达的影响。②方法 高三尖杉酯碱(H)、阿糖胞苷(A)、足叶乙甙(V)3种化疗药物分别作用于AML患者的单个核细胞,一组加HBO,另一组不加HBO,观察各组凋亡细胞、Bax和bcl-2实验前后积分变化、Bax/bcl-2积分比和TUNEL阳性率。同时设无药加HBO组和不加HBO组以排除HBO或其它因素对AMI,细胞的影响。③结果 3种化疗药物加HBO组具有典型凋亡特征的AML细胞明显增多;Bax的表达无明显改变(P>0.05),bcl-2的表达明显减少(P<0.01),Bax/bcl-2积分比明显高于不加HBO组(P<0.05);TUNEL 阳性率明显高于不加HBO组(P<0.01);无药加HBO组与无药不加HBO组实验前后Bax、bcl-2基因的表达无明显改变(P>0.05)。④结论HBO对化疗药物体外促进AML细胞Bax基因的表达无明显影响,但增强了化疗药物对bcl-2基因表达的抑制作用,Bax/bcl-2比值明显增加,因而增强了AML细胞的化疗敏感性,使化疗药物体外诱导AML细胞凋亡增加。单纯HBO对AML细胞Bax和bcl-2基因的表达无明显影响。

Abstract:Objective To investigate the influence of hyperbaric oxygen(HBO) to chemotherapeutic drugs inducing the expression of gene Bax and bcl-2 in acute myeloid leukemia (AMD cells in vitro. Methods 3 chemotherapeutic drugs(HHT,Ara-c,VP-16) were treated to mononuclear cells of AML which were divided into two groups (one with HBO and the controls without HBO). Apoptotic cells of two groups were observed and the changes of Bax and bcl-2, Bax/bcl-2 ratios and TUNEL positivity rates of two groups were also detected. At the same time,AML cells were also divided into two groups,one with HBO and the controls without HBO in order to avoid the influence of other factors. Results There were more apoptotic cells in the groups with HBO of three drugs. The expression of gene Bax had no obvious changes(P>0. 05) while the expression of gene bcl-2 obviously decreased(P<0. 01). The groups with HBO of three drugs also had higher Bax/bcl-2 ratios (P<0. 05) and higher TUNEL positivity rates (P<0. 05) contrast to the controls. Conclusion HBO had no influence to the expression of gene Bax in vitro,but it could enhance the effect of chemotherapeutic drugs inducing the expression of gene bcl-2 in AML cells in vitro . HBO might enhance the sensitivity of AML cells to chemotherapeutic drugs by depressing the expression of gene bcl-2 and could enhance chemotherapeutic drugs inducing AML cells apotosis in vitro.
