结肠癌组织中maspin及uPA基因的表达与临床意义

摘    要:目的通过对结肠癌及正常结肠黏膜组织标本中maspin和uPA基因的表达进行检测,探讨二者与结肠癌病理学特征之间的关系,以及在组织中表达的相关性,为临床寻找结肠癌早期诊断指标提供理论依据。方法采用免疫组织化学SP法测定62例结肠癌及30例正常结肠组织中maspin以及uPA基因的表达,对结果进行分析。结果 maspin蛋白的阳性表达率在结肠癌未侵及浆膜、淋巴结无转移及TNM分期0~Ⅰ期者中明显高于侵及浆膜、淋巴结有转移及TNM分期Ⅱ~Ⅳ期者(P均〈0.05),在结肠癌不同性别患者中无差异(P〉0.05)。uPA蛋白的阳性表达率在结肠癌未侵及浆膜、淋巴结无转移及TNM分期0~Ⅰ期者中明显低于侵及浆膜、淋巴结有转移及TNM分期Ⅲ~Ⅳ期者(P均〈0.05),在结肠癌不同性别患者中亦无差异(P〉0.05)。maspin与uPA基因在结肠癌的表达中呈负相关(rs=-0.416,P〈0.05)。结论 maspin和uPA基因在结肠癌和正常结肠黏膜组织中的表达有明显差异,且与结肠癌的病理学特征相关,二者在结肠癌组织样本中的表达也存在相关性,可作为反映结肠癌病理生物学行为的指标。

Abstract:Objective To investigate the expression of maspin and urokinase-type plasminogen activator(uPA) gene in the mucosa of colon cancer and their roles in the tumorigenesis and progression of colon cancer.Methods Immunohistochemistry SP method was used to detect the expressions of maspin and uPA gene in mucous tissues from colon cancer(n=62) and normal colon epithelium(n=30).Results The positive expression rates of maspin protein in colon cancer with non-serosal invasion,without lymph node metastasis and with TNM stages 0-1 were significantly higher than those in colon cancer with serosal invasion,lymph node metastasis and TNM stages Ⅱ-Ⅳ(all P0.05),whereas the positive expression rates of uPA protein in colon cancer with non-serosal invasion,without lymph node metastasis and with TNM stages 0-1 were significantly lower than those in colon cancer with serosal invasion,lymph node metastasis and TNM stages Ⅱ-Ⅳ(all P0.05).There were no gender differences in positive expression rate of maspin protein and uPA protein.Maspin gene expression had a negative correlation with uPA gene expression in the tissues of colon cancer(rs=-0.416,P0.05).Conclusions There is a significant difference on the expressions of maspin gene and uPA gene in mucous tissues of the colon cancer and normal colon.Down-regulated expression of maspin and up-regulated expression of uPA might play important roles in the invasion and metastasis of colon cancer.They also present a correlation in expression of different stage tissue of colon cancer.Therefore,they may serve as the effective biomarkers reflecting pathology of colon cancer.
