柴芍承气汤加味银杏叶对重症急性胰腺炎大鼠血PAF、TNF-α、IL-8的影响

摘    要:目的通过动物实验方法探讨柴芍承气汤加味银杏叶对重症急性胰腺炎(SAP)大鼠模型胰腺损伤及血中PAF、TNF-α、IL-8水平的影响。方法将72只成年雄性Wistar大鼠随机分为4组:假手术组(J组)、SAP模型组(M组)、柴芍承气汤组(C组)、加味柴芍承气汤组(Z组),各组又按制模后规定时间分为6、12、24h3个亚组,每组6只。采用牛黄胆酸钠胰胆管注射法制备SAP大鼠模型,J组大鼠仅于剖腹手术后轻轻翻动胃与十二指肠。制模后,C组给予柴芍承气汤灌胃,Z组给予柴芍承气汤加味银杏叶鼻饲,J组与M组给予生理盐水灌胃。大鼠处死后取胰腺组织,标本用10%福尔马林固定,石蜡包埋,切片,HE染色,光镜观察各组胰腺组织损伤情况。制模后,按规定时间点取大鼠动脉血,分别测定各组血中PAF、TNF-α、IL-8水平变化。结果 M组胰腺组织损伤随着时间延长逐渐加重,血中PAF、TNF-α、IL-8水平逐渐增高;C组较M组胰腺组织损伤减轻,血中PAF、TNF-α、IL-8水平均降低;Z组较C组血PAF水平下降。结论柴芍承气汤加味银杏叶可通过降低SAP大鼠血TNF-α、IL-8和PAF水平,对胰腺组织起到保护作用,其较柴芍承气汤具有更好的治疗作用。

Abstract:Objective To explore the impact of Chaishao Chengqi decoction with ginkgo biloba leaf(GBL) on damage of pancreas tissue and the levels of serum platelet activating factor (PAF),tumor necrosis factor-α(TNF-α) and interleukin-8 (IL-8) in rats with severe acute pancreatitis (SAP).Methods Seventy-two Wistar male rats were randomly divided into four groups:sham operation group(J group),SAP model group (M group),Chaishao Chengqi decoction group (C group) and combination of Chaishao Chengqi decoction and GBL (Z group),and each group (n=18) was further divided into three subgroups based on the time points of constructing model:6 h,12 h and 24 h subgroups (n=6 each).The model of SAP rat was constructed through injecting sodium taurocholate into pancreatic and biliary duct.In the rats of J group,the duodenum and pancreas were only turned over gently after laparotomy.After constructing model,the Chaishao Chengqi decoction was perfused into the stomach in C group;the Chaishao Chengqi decoction with GBL was given by nasal feeding in Z group;0.9% NaCl was perfused into the stomach in J and M groups.The samples of pancreas and lung were taken after killing experimental rats.The samples were disposed:fixation by 10%formalin,paraffin embedding,sectioning and HE staining.The samples were observed under optic microscope.Arterial blood of rats was taken in the fixed times,and serum PAF,TNF-α and IL-8 were detected by ELISA method.Results With the prolongation of times,The damage of pancreatic tissue gradually exacerbated,and the levels of PAF,TNF-α and IL-8 gradually elevated in the M group.Compared with M group,the damage of pancreatic tissue alleviated,and the levels of PAF,TNF-α and IL-8 descended in the C group.Compared with C group,the level of PAF descended in group Z.Conclusions Chaishao Chengqi decoction with GBL could play a protective role to pancreatic tissue probably via lowering the levels of serum TNF-α,IL-8 and PAF,and its therapeutic effect is superior to Chaishao Chengqi decoction alone.
