| 摘 要： | 建立SARS-CoV病毒分离培养方法,确定SARS-CoV病毒感染动物后在不同时间和不同组织病毒的复制和存活时间,并比较中国恒河猴和食蟹猴对病毒的敏感性,筛选出对SARS冠状病毒敏感的动物,完善SARS冠状病毒感染猴模型的病毒学指标。方法:SARS-CoV经鼻腔接种实验用猴,定期采集动物的鼻咽拭子、血浆和组织标本进行病毒分离培养,分离出来的病毒用免疫荧光方法和中和试验等方法进行测定。结果:SARS冠状病毒均可感染恒河猴和食蟹猴,在鼻咽拭子、血浆和组织中可以分离到病毒。结论恒河猴和食蟹猴均可作为SARS感染动物模型,而病毒分离培养可以作为感染模型的重要指标。

| Abstract: | Objective:To establish the method for isolation of SARS-coronavirus(SARS-CoV),try to confirm whether rhesus or cynomolgus macaques could be infected with SARS-CoV,and determine the distribution and duration of the virus animals as SARS model.Method The monkeys were infected with identified SARS-CoV isolated from human SARS patient.The swab and serum samples were collected at 4,7,10 days after inoculation.Tissues were collected at different time.All samples were cultured with Vero-E6 cells.Result The SARS-CoV was detected from various samples from infected animals.Conclusion Viral isolation is an unreplaced method for testing virus replication in vivo |
