人组织型激肽释放酶基因对2型糖尿病大鼠AMP激活的蛋白激酶活性的影响

摘    要:目的 研究人组织型激肽释放酶(human tissue kallikrein, HK)基因对链脲佐菌素(streptozotocin,STZ)诱导的2型糖尿病(diabete mellitus, DM)大鼠AMP激活的蛋白激酶(AMP-activated protein kinase, AMPK)活性的影响.方法 48只雄性Wistar大鼠随机分为普通饲料组,即正常对照组(n=16),和高脂高糖饲料组(n=32),高脂高糖饲料组大鼠以小剂量STZ加高脂高糖饮食诱导2型DM,再将其随机分为两组,分别经尾静脉导入重组腺相关病毒(recombinant adeno-associated viruses, rAAV)介导的HK基因(HK组)以及报告基因beta-半乳糖苷酶(beta-galactosidase, Lacz)(Lacz组),观察12周后处死实验动物,留取心脏、肝脏、肾脏等组织.Western blot法检测HK在各脏器的表达以及AMPK的表达及其磷酸化水平(phospho-AMPK).结果 rAAV介导的HK可在2型DM大鼠的肝脏、肾脏、心脏组织表达,且HK组各脏器中HK蛋白表达显著高于Lacz组(均P<0.05).Western blot检测发现,在肝脏、肾脏、心脏、骨骼肌、脂肪等脏器中,HK组和Lacz组AMPK的表达与正常对照组比较,差异无显著性意义(均P>0.05),而HK组phospho-AMPK较正常对照组显著上调(P<0.05),Lacz组phospho-AMPK与正常对照组比较,差异无显著性意义(P>0.05).结论 rAAV载体介导的HK基因能够激活2型DM大鼠主要器官AMPK的表达.
