摘    要:介绍两种简便可行的EB病毒转化B淋巴细胞的方法,即微量全血法和冻存全血法。与环孢霉素A法相比较,这两种方法具有操作简单,所需血样少,无需环孢霉素A等优点。这些特点尤其是在进行大量样本转化时显得更为突出。用微量全血法和冻存全血法共进行79例转化试验,转化成功率分别为46%与85%。对于影响这两种EB病毒转化结果的因素也进行了讨论。

Abstract:Two simple practical methods to transform human B lymphocyte into B lymphoblastoid cell lines (LCL) by Epstein-Barr virus (EBV) are reported in this paper. In these methods only small amount of fresh whole blood and small amount of frozen whole blood were used. The 79 samples were trans formed by EBV using the two methods, and the successful rates were 46% and 85% respectively. Compared with the classical Cyclosporin A method, the two methods were simple and practical, with only a small amount of blood required, and no special reagents needed. These characteristics are more important when large numbers of samples are to be handled. In addition, the factors affecting the successful transformation were discussed in this paper.
