摘    要:目的构建能合成和分泌γ-氨基丁酸(γ-aminobutyric acid,GABA)且可以无限增殖的永生化神经前体细胞。方法在永生化的神经前体细胞中转人含有GABA合成限速酶谷氨酸脱羧酶65亚型(glutamate decarboxylase 65,GAD65)的基因,用免疫组织化学、Western blot、毛细管电泳等方法观察GABA的合成和分泌。结果与对照组相比,INPCs—GAD65的含量明显增加。合成和释放的GABA显著增加,并能记录到GABA电流。结论成功构建了能合成和分泌GABA的永生化的神经前体细胞。

Abstract:Objective To construct genetically engineered cell lines to synthesize and release GABA by immortalized neural progenitor cells(INPCs) in rats.Methods The INPCs in rats were transfected with the GABA-synthesizing enzyme GAD(65) cDNA(INPCs-GAD65).The synthesis and secretion of GABA was observed by capillary electropherograms and the expression of GAD65 detected by immunohisochemistry and Western blot,respectively.GABA current was recorded in INPCs using whole-cell voltage clamp technique.Results In INPCs-GAD65,the contents of GAD65 were significantly increased as compared with control group,GABA were synthesized and secreted,and the GABA current was recorded.Conclusion The INPCs-GAD65 synthesizing and secreting GABA were successfully constructed.
