摘    要:目的检测促血管生成素1(Ang1)蛋白在小鼠胚胎着床期子宫内膜的分布及mRNA的表达,以探讨其在胚胎着床过程中所起的作用和生物学意义。方法取妊娠2、4、6和8d的小鼠子宫内(蜕)膜,分别运用免疫组织化学SP法和原位杂交技术,检查着床期子宫内膜中Ang1蛋白的表达及其mRNA的转录水平。并用计算机图像分析技术检测不同时期子宫内膜中Ang1表达的平均吸光度值。结果Ang1特异性免疫反应产物在基质细胞中表达,随着妊娠天数的增加,表达逐渐增强(P<0.01),且上皮细胞和血管壁亦为Ang1特异性免疫反应阳性结构。Ang1mRNA自妊娠第2天起即表达于基质细胞中,第4天血管壁细胞质中也出现阳性表达,且两者的表达强度在妊娠第6、第8天时逐渐增加,平均吸光度值差异均有显著性意义(均P<0.01)。结论Ang1基因在胚胎着床期血管新生和血管重塑过程中起重要作用,有助于囊胚成功着床。

Abstract:Objective To investigate the expression and distribution of angiopoietin-1 (Ang-1) in the endometrium of early pregnancy mice.Methods The expression of Ang-1 was detected by immunohistochemical staining and in situ hybridization methods respectively. Image analysis system was used to measure the average absorbancy of Ang-1 in endometrium of different stages of pregnancy and the controls.Results The expression of Ang-1 was markedly different among the four groups. Immunohistochemistry revealed Ang-1 localized in the cytoplasma of stromal cells on the 2nd day of pregnancy and in luminal epidermal cells on the 4th day respectively. The expression intensity of Ang-1 was gradually increased in 2-, 4-, 6- and 8-day pregnancy mouse endometrium (P<0.01). In situ hybridization indicated that Ang-1 mRNA was detected in stromal cells at 2nd day of pregnancy, hybridization signals localized in both stromal cells and vascular epithelial cells on the 4th day, and the positive expression was increased on the 6th day and 8th day (P<0.01).Conclusion Ang-1 may play an important role in angiogenesis and vascular remodeling of endometrium during the process of embryo implantation.
