摘    要:目的 :通过化学萃取同种异体神经 ,去除髓鞘和雪旺细胞 ,形成无细胞基膜管后桥接鼠坐骨神经缺损 ,研究神经再生效果。方法 :正常鼠坐骨神经用非变性生物剂处理后得到无细胞的基膜管 ,桥接鼠坐骨神经 2 0mm缺损。实验分 3组 :无细胞基膜管移植组 (A组 )、自体神经移植组 (B组 )和异体神经移植组 (C组 )。术后进行肌电图、组织形态学及图像分析仪检查。结果 :A组再生神经有大量轴突通过移植体 ,术后 2个月电生理检测再生神经的潜伏期及波幅低于B组 (P <0 .0 5 ) ,3个月时差异无显著性。髓鞘厚度在术后 3个月时亦低于B组 ,差异有显著性 (P <0 .0 5 )。轴突直径及数目两组无差异。结论 :这种无细胞基膜管移植体能支持轴突的生长和雪旺细胞的迁移 ,是一种良好的神经移植替代材料。

Abstract:Objective: To study the effect of nerve regeneration through bridging the gap of rat sciatic nerve with acellular basal laminea tubes which Schwann cell and myelin sheath was removed. Methods: Acellular basal laminea tubes were acquired from normal rat sciatic nerves which were treated with nondenaturing biological detergents. Thirty adult SD rats were chosen as experimental model and 20 mm length of left sciatic nerve was cut and repaired. They were divided randomly into three groups(10 SD rats in each group):group of acellular basal lamina tubes graft(group A),group of autogenous nerve bridging(group B)and group of allogenic nerve bridging(group C). A gap of 20 mm of rat left sciatic nerve was extracted,which was bridged by different nerve materials. Histomorphometry, electromyographic and image analysis was determined 2 and 3 months post operation. Results: A number of axons in regenerating nerve were found to cross the graft in group A,and the latency and wave amplitude in electromyograph of regenerating nerve in group A was lower than that in group B(P<0.05)at the second month,there was no statistical difference in both group A and B at the third month. The thickness of myelin sheath of experimental group was inferior to that of group B(P<0.05)at the third month,while the number and diameter of axons in both groups was similar. Conclusion: The grafts support outgrowth of axons and migration of Schwann cell,and is valuable in peripheral nerve regeneration.
