归芪口服液抗大鼠肾间质纤维化的实验研究

摘    要:目的:研究单侧输尿管梗阻后肾间质纤维化的发生机理及中药归芪口服液对其保护作用。方法:将24只大鼠随机分假手术组即对照组、手术组和归芪治疗组,采用单侧输尿管梗阻(UUO)模型,术后第8d观察梗阻肾组织病理改变,应用免疫组织化学方法检测转化生长因子β1(TGF-β1)和α-平滑肌肌动蛋白(α-SMA)的表达,并应用放射免疫法检测血浆和肾组织血管紧张素Ⅱ(AngⅡ)的含量。结果:手术组肾组织TGF-β1、α-SMA的表达增加,血浆和肾组织AngⅡ增高,与对照组相比有显著差异(P<0.01);归芪治疗组与手术组相比,TGF-β1、α-SMA的表达下调,AngⅡ下降,差异有显著性(P<0.05)。结论:归芪口服液可能通过抑制AngⅡ的生成,下调TGF-β1、α-SMA的表达而阻抑肾间质纤维化的进展。

An Experimental Study of Guiqi Oral Liquid on Inhibition of Renal Interstitial Fibrosis in Rats

Abstract:Objective: To investigate the mechanism of renal interstitial fibrosis after unilateral ureteral obstruction and the renoprotective effect of Guiqi oral liquid, a traditional Chinese medicine. Methods: Twenty four rats were randomly assigned to the control group (sham operation group), operation group and Guiqi treatment group after the unilateral ureteral obstruction. Renal tissues were examined by light microscopy two weeks after operation. Immunohistochemistry was applied to measure the expression of transforming growth factor-β_1(TGF-β_1) and α-smooth muscular actin (α-SMA). Radio-immunoassay was used to quantitate level of angiotensinⅡin renal and plasma. Results: In comparison with that in the control group, the expression of TGF-β_1 and α-SMA in the interstitium in the operation group was significantly increased(P<0.01). The levels of angiotensin Ⅱ in renal and plasma in the operation group were significantly increased too (P<0.01). In comparison with operation group, Guiqi oral liquid could significantly decrease the expression of TGF-β_1 and α-SMA (P<0.05), and could significantly reduce the level of angiotensinⅡ(P<0.05). Conclusion: Guiqi oral liquid may protect renal by inhibiting the overexpression of TGF-β_1,α-SMA and reducing the level of angiotensinⅡ in renal and plasma.
