用基因芯片技术检测宫颈癌患者相关基因的差异表达

目的:研究感染HPV18的宫颈癌组织中差异表达的基因,并进一步探讨HPV18致宫颈癌发生的机制。方法:采用杂交捕获Ⅱ代技术筛选出3例感染HPV18的宫颈癌病例,取癌组织及自身正常宫颈组织标本,用含14 112条人类全长基因的cDNA微阵列芯片筛选出差异表达的基因并进行分析。结果:在3例宫颈癌组织中共同差异表达的基因有1 315条,其中649条上调,666条下调。这些基因涉及原癌基因和抑癌基因、细胞周期蛋白类、细胞信号和传递蛋白、代谢、免疫相关、细胞受体、蛋白翻译合成以及其它一些未知功能的基因。结论:HPV18引发宫颈癌是多因素多基因共同作用的结果,它们通过相互调节、网络调控直接或间接地导致宫颈癌的发生发展,可望进一步分析证实以明确HPV引发宫颈癌的精确分子机制。

Differential Gene Expression Profile in Cervical Cancer Infected with Human Papillomavirus 18 Screened out by cDNA Microarray

Objective: To indentify a set of genes related to human papillomavirus (HPV) of cervical cancer and to discuss the pathogenesis of cervical cancer.Methods: Three women of cervical cancer infected with HPV-18 were selected by Hybrid capture Ⅱ method.The human gene expression of cervical cancers and their normal cervical tissues was analyzed with 14 112 human genes of cDNA microarrays.Results: A total of 1 315 differential genes expressed in the cervical cancer were screened out,of which,649 were up-regulated and 666 were down-regulated.These genes involved in the signal transduction,metabolism,immunity response,protein biosynthesis and ESTs.Conclusion: Cervical cancer induced by HPV18 is a result of co-effecting by polygene and multiple factors.These genes were regulated by each other,play roles directly or indirectly in the carcinogenesis of cervical cancer.And cDNA microarray technique is effective in screening out the differentially expressed genes for the further investigation.
