Detection of E2 Gene Deletion in the Evaluation of HPV16 Infection in Cervical Lesions

Objective: To investigate the relationship between cervical cancer and the presence of HPV 16 viral chromosome in cervical lesions by application of E2 gene deletion assay.Methods: HPV-E6 gene as well as C-terminal,N-terminal and hinge region of HPV16 E2 from specimens of patients infected with HPV16 were amplified by multiple polymerase chain reaction(PCR).Scion Image 4.0 electrophoresis image analysis soft was used to evaluate the area of electrophoresis products of E2 gene and E6 genes for the analysis of the integration state of HPV16.Results: The HPV16 infection percentage was 67.14% and 38.10% respectively in 70 cases of cervical cancer group and 40 cases of non-cancerous benign lesions group.In the cervical cancer group,the HPV16 genome of 40 cases was in pure integrated form,6 cases in the mixture of episomal and integrated forms,and one case in the pure episomal form.In the non-cancerous cervical lesions group,5 cases was in the pure integrated form,one case in mixture of episomal and integrated forms,and 10 cases in pure episomal form.Episomal states existed in all the normal cervical tissue samples.Conclusion: There are close links between E2 gene deletion and HPV16 infection.For the patients with cervical lesions,the integration status of HPV 16 can be used to analyze the prognosis and help to choose the proper treatment.
