Establishment of an Animal Model of Periodontitis Using the Old Rat of Kidney Deficiency Physique

Objective: To build an experimental animal model of periodontitis with kidney deficienly physique according to the theory of traditional Chinese medicine,in order to study the periodontitis treatments by integrated traditional and western medicine. Methods: A total of 40 young (4 months old) and 40 aged (12 months old) female or male SD rats were randomly divided into four groups: the young normal group(YN, n=20), the young periodontitis group(YP, n=20), the old normal group(ON, n=20) and the old periodontitis group(OP, n=20).YN and ON groups were anaesthetized by 100 mg/kg ketamine intraperitoneal injection and fed with regular diet.YP and OP groups were anaesthetized by 100 mg/kg ketamine intraperitoneal injection, treated with 0.2 mm orthodontic steel ligation in the neck of the first molar, cemented with GC, and fed with high sugar diet. Results: Eight weeks after operation, histopathologic examination revealed that the periodontal tissue of YN and ON group had no pathological changes.YP group showed a periodontal pocket, a large number of osteoclasts infiltration and alveolar ridge top absorption.OP group showed a periodontal pocket, alveolar ridge top absorption and alveolibone loss 1/3 part to root apex. Conclusion: Aged SD rats have a higher susceptibility than young rats. Periodontitis rat models of kindney deficiency physique were successfully established and laid the foundation for treating it with integrated traditional and western medicine.
