缺氧诱导因子和血管生成素在直肠癌中的表达及血管形成中的作用

摘    要:目的:研究缺氧诱导因子(HIF-1α)及血管生成素(Ang-2)在直肠癌组织中的表达,及其与临床病理特征和肿瘤新生血管形成之间的关系。方法:采用免疫组化法检测56例直肠癌组织和20例切缘正常组织中HIF-1α、Ang-2和CD105的表达。结果:HIF-1α和Ang-2在直肠癌组织和切缘正常组织中的表达差异有统计学意义(P<0.05);HIF-1α和Ang-2在直肠癌组织中的表达均与直肠癌的临床分期和血行转移有关(P<0.05);而与患者的性别、年龄、淋巴转移和浸润深度无关(P>0.05),Ang-2在直肠癌组织中的表达与肿瘤大小、分化程度有关(P<0.05),并且在溃疡型和隆起型中阳性表达的差异有统计学意义(P<0.05)。HIF-1α、Ang-2阳性表达的直肠癌组织中微血管密度(MVD)数值比阴性表达组显著增高(P<0.05),且三者的表达两两之间呈正相关(r=0.333,r=0.355,r=0.280;P<0.05)。结论:HIF-1α和Ang-2在直肠癌中的高表达可能与预后、转移以及肿瘤新生血管形成有密切关系。

Abstract:Objective: To investigate the relationship between the expression of Hypoxia-Inducible Factor-1α(HIF-1α) and angiopoietin-2(Ang-2) proteins and clinical characteristics in human rectal carcinoma,and to explore the relation between HIF-1α or Ang-2 and oncogenesis.Methods: Immunohistochemical technique was used to detect the expression of HIF-1α,Ang-2 and CD105 protein in 56 rectal carcinoma and 20 adjacent normal tissues.Results: The positive rates of HIF-1α and Ang-2 were significantly higher in rectal carcinoma than those in normal tissues(P<0.05).The expressions of HIF-1α and Ang-2 were related with Duke's stage and the distal metastasis(P<0.05),but had no marked correlations with the sex,age,lymph node metastasis or invasion depth(all P>0.05).The level of Ang-2 expression was also correlated with the size and histological differentiation of rectal carcinoma,and the positive rate of Ang-2 was different between ulcer type and protrude type.The MVD in HIF-1α or Ang-2 positive group was significantly higher than in HIF-1α or Ang-2 negative group.There were significant correlations among the expression of HIF-1α,Ang-2 and CD105 proteins.Conclusion: High expression of HIF-1α and Ang-2 proteins in rectal carcinoma may play an important role in the progression,metastasis and oncogenesis.
