摘    要:目的 :探讨尿内皮素 - 1(ET - 1)在大鼠药物性肾损害中的变化及意义。方法 :采用庆大霉素制造动物模型、观察庆大霉素致大鼠急性肾损伤的表现 ,观察尿ET - 1、尿糖、尿钠、2 4h尿蛋白定量等指标变化。结果 :肾损伤组尿ET - 1含量进行性升高 ,与正常对照组差异有显著性 (P <0 .0 1)。结论 :尿ET - 1参与了肾毒性肾损伤的发生与发展 ,尿ET - 1含量进行性升高显示肾损伤程度加重。

Abstract:Objective: To investigate the correlation of changes of plasma and urinary endothelin(ET) concentrations in rats with gentamycin-induced nephrotoxicity. Methods: 20 SD rats were randomly divided into control and nephrotic group. The nephrotic animal model was made by a intraperitoneal injection of gentamycin. At the second week, the excretion of 24 hours urinary protein, urinary glucose and Na concentrations were assayed by biochemistry methods. Plasma and urinary endothelin (ET) concentrations at the first and second week were also measured by means of radioimmunoassay(RIA). Results: The excretion of 24 h urinary protein, urinary glucose and Na , plasma and urinary endothelin in nephrotic group were significantly higher than those in control group (all P<0.01). Conclusion: ET-1 may be involved in the development and progression of gentamycin-induced nephrotoxicity and the up-regulation of urinary ET-1 may be used as a predictor for it.
